Is the Nightwarden required?

No. The Nightwarden’s quest, “Your test begins.”, is marked optional, and the quest that opens Act V only needs End Highlands, which comes right after the Ender Dragon.

The fight is still worth it later: it gives 250 XP levels, two Universium Ingots, two Perfect Gems and a Wisdom Scroll. See Defeat The Nightwarden.
